Allow two hours, get the audio tour in your language, and go before you go to visit the temples. It provides a useful foundation of historical and cultural information that will deepen your experience of the temples and Siem Reap. It is easy to get a Tuk-tuk or Remork to and from the museum.
I recommend a thorough exxperience of this museum BEFORE you visit any of the ruins. Knowing and understanding what you'll be looking at makes all the difference in the world!
